If it doesn't affect his teaching abilities, then it doesn't matter, does it? What you look like has no relation to whether or not you are skilled at conveying concepts to people in order for them to learn.
When I think back to my schooling, and I think through my excellent teachers and my dreadful ones, sexuality never really came into things. (Apart from my biology teacher who had the most magnificent rack, and my senior art teacher who was an absolute babe in the Helena Bonham Carter, Bill and Ted's Excellent adventure sense)

You don't need to test for their Y chromosome, as few, if any, male homosexuals deny that they are physically male. They are not confused about their gender, you are confused about their sexuality.
I could picture something like this actually affecting his teaching abilities, if only because the students probably won't be very mature about it if they find out. That's more a sad commentary on the state of our society than it is on the teacher himself, but it still may be the case, unfortunately.

If it isn't affecting their ability to teach, then someone's gender identity is nobody else's damn business. Good teachers are hard to find, and this is particularly true in areas where Fundie idiots hold the most sway, because smart people get the fuck out. But I imagine that suits people like Mr. Knight just fine.
So, let´s imagine this scenario. If I undergo surgical operation because I don´t feel well with my aspect, am I less qualified as a teacher?, or if I convert to another religion, or change degree?
