All that is on Plum Island is an agricultural research station. It is highly secure because the government is experimenting with hoof and mouth disease there and want to keep it quarantined there and not get into the US livestock population.

I doubt that CIA would need to create a "threat" that has already been created. This conspiracy theory of yours is flimsy and far too risky/inefficient for modern-day intelligence agencies. Besides, drug-resistant strains aren't exactly a new thing and Tuberculosis is pretty shitty for propaganda purposes and as a bio-weapon.

All it took for a completely drug-resistant strain of TB to surface is a malnourished population, living in crowded, unhygienic conditions, who because of inadequate access to medical care and treatment resort to self- help with insufficient antibiotics, and all it takes to spread it around the world is some jet-setters and a lot of uninspected produce. This is, in short, the fruit of so-called free enterprise and its increasingly two-class society, and the CIA doesn't have a damn thing to do with it.

You do appreciate that drug-resistant TB shows up mostly outside the US, right? And that the completely resistant strain has only been found in India and possibly in Iran so far?

The US isn't to blame for this one, except to the extent that dedicated campaigns to contain multi-resistant TB, which this one evolved from, could have happened much earlier than they did if there had been more investment and then we might not have had the resistant bugs crossing with each other to make the this new strain. And the US far from the only culprit there.
